Description

TEV protease is a site-specific endoprotease that cleaves after glutamine at its cleavage site Glu-Asn-Leu-Tyr-Phe-Gln-/-Gly. Recombinant TEV protease is purified from E.coli containing a clone of the Tobacco Etch Virus gene. Since TEV protease contains 6x His sequence (EBF-1009 and 1010) or 6x His-GST (EBF-1011 and 1012), they can be easily removed by a metal affinity chromatography or GSH affinity chromatography after the cleavage reaction. The recombinant 6x His tagged TEV protease is 28.6 kDa, and 6x His-GST tagged TEV protease is 58.1 kDa in size.

Product Component

- TEV Protease : 100 μl (100 mg, 2,000 units), or 500 μl (500 mg, 10,000 units)

- Control protein : 30 mg (1 mg/ml), produced 45 kDa and 48 kDa by TEV protease

Unit Definition

One unit of TEV cleaves ≥95% of 3 μg control substrate in 1 hr at 30°C.

Specific Activity

TEV Protease (6x His Tag) : 20 unit/mg protein

TEV Protease (6x His, GST Tag) : 10 unit/mg protein

Storage Buffer

50 mM Tris-HCl, pH 7.5, 1 mM EDTA, 5 mM DTT, 50% glycerol, 0.1% Triton X-100.

TEV Reaction Condition

50 mM Tris-HCl, pH 8.0, 0.5  mM EDTA, 1 mM DTT, 150 mM NaCl.
