Description

T4 RNA Ligase catalyzes the ATP-dependent ligation of single-stranded DNA or RNA onto the 5´-phosphoryl termini of single-stranded DNA or RNA. T4 RNA Ligase also catalyzes the addition of [5´-32P] nucleoside 3´,5´-bis (phosphate) onto single-stranded RNA. T4 RNA Ligase is purified from a recombinant E.coli strain.

Concentration & Storage Condition

10 unit/ml.  Store at -20℃.

Storage Buffer  

10 mM Tris-HCl, pH 7.5, 50 mM KCl, 0.1 mM EDTA, 1 mM DTT, 0.1% Tween 20, 50% glycerol.

10x Reaction Buffer

500 mM Tris-HCl, pH 7.8, 100 mM MgCl2, 50 mM DTT, 10 mM ATP.

Unit Definition  

One unit catalyzes the formation of 1 nmole of [5´-32P]rA14–20 into a phosphatase-resistant form in 30 min at 37°C in a total volume of 30 ml. The reaction conditions are: 50 mM Tris-HCl, pH 7.8, 10 mM MgCl2, 5 mM DTT, 1 mM ATP and 10 mM [5´-32P]rA14–20.
