The Hind III methylase recognizes 5´--AAGCTT--3´ sequences and methylates first adenine (N⁶) residues.
Source- Recombinant gene from Haemophilus influenzae Rd.Applications- Blocking restriction endonuclease cleavage restricted by Hind III methylation.Reaction Conditions- 1x Methylase reaction buffer in the presence of 160 μM S-adenosylmethionine (SAM). Incubate at 37℃.Concentration & Storage Condition- Store at -20℃. 5 unit/㎕ in 50 mM Tris-HCl, pH 7.5, 50 mM KCl, 1 mM DTT, 10 mM EDTA, 200 ㎍/ml BSA, 50% (v/v) glycerol.Unit Definition- One unit is defined as the amount of enzyme required to fully protect against Hind III cleavage of 1 ㎍ of λ DNA in 1 hr at 37℃.Heat Inactivation Condition- 65℃ for 20 min.QC Tests
