The Hpa II methylase recognizes 5´--CCGG--3´ sequences and methylates internal cytosine (C⁵) residues.
Source - Recombinant gene from Haemophilus parainfluenzae. Applications- Blocking restriction endonuclease cleavage restricted by Hpa II methylation. Reaction Conditions- 1x Methylase reaction buffer in the presence of 160 μM S-adenosylmethionine (SAM). Incubate at 37℃. Concentration & Storage Condition- Store at -20℃. 10 unit/㎕ in 50 mM Tris-HCl, pH 7.5, 100 mM NaCl, 1 mM DTT, 10 mM EDTA, 200 ㎍/ml BSA, 50% (v/v) glycerol. Unit Definition- One unit is defined as the amount of enzyme required to fully protect against Hpa II cleavage of 1 ㎍ of λ DNA in 1 hr at 37℃. Heat Inactivation Condition - 65℃ for 20 min. QC Tests
