tRecA protein is necessary for genetic recombination events of thermostable bacteria, Thermus thermophilus. tRecA promotes the strand exchange of single-strand DNA fragments with homologous duplex DNA. The reaction has three distinct steps : (1) tRecA polymerizes on the single-strand DNA, (2) the nucleoprotein filament binds the duplex DNA and searches for a homologous region, (3) the strands are exchanged. tRecA is purified from E. coli strain carrying an over-expressed tRecA gene. tRecA is thermostable protein and can be applied to PCR reaction to improve sequence specificity.
